Fig.1 Detection of RHBDF2 expression in lung by RT-PCR. Lung RNA was extracted from 6-week-old female wild-type C57BL/6 (WT)(n=2) and male homozygous hRHBDF2 knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Rhbdf2 mRNA (452 bp) was detectable only in wild-type C57BL/6 mice. Human RHBDF2 mRNA (571 bp) was detectable only in homozygous hRHBDF2 knockin mice but not in wild-type mice. 

Fig.2 Detection of RHBDF2 expression in hRHBDF2 mice by WB. The spleen, lung, thymus and bone marrow tissue lysates were collected from 6-week-old female wild-type C57BL/6 mice and 6-week-old male homozygous hRHBDF2 mice, and then analyzed by western blot with anti-RHBDF2 antibody. RHBDF2 was detectable in spleen, lung, thymus and bone marrow from both WT C57BL/6 and HO hRHBDF2 mice , as the antibody was cross-reactived between human and mouse RHBDF2.
