Fig.1 Detection of PNPLA3 expression in liver by RT-PCR. Liver RNA was extracted from 7-year-old male wild-type C57BL/6 (WT)(n=1) and homozygous hPNPLA3-I148M(2) knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Pnpla3 mRNA (341 bp) was detectable only in wild-type C57BL/6 mice. Human PNPLA3 mRNA (343 bp) was detectable only in homozygous hPNPLA3-I148M(2) knockin mice but not in wild-type mice. 

Fig.2 Detection of PNPLA3 expression in hPNPLA3-I148M(2) mice by WB. The liver tissue lysates were collected from 7-week-old male wild-type C57BL/6 mice and 7-week-old homozygous hPNPLA3-I148M(2) mice, and then analyzed by western blot with anti-PNPLA3 antibody. PNPLA3 was detectable in liver from both WT C57BL/6 and HO hPNPLA3-I148M(2) mice , as the antibody was cross-reactived between human and mouse PNPLA3.
